چکیده

A variety of nucleoside analogs have shown carcinostatic activity. In some instances they are relatively rapidly excreted. In others they are relatively rapidly inactivated in vivo by catabolic enzymes. In both cases it appeared desirable to have nucleoside derivatives that would act as depot forms of the nucleoside analogs and would be converted in vivo to the respective nucleoside analog. Gish et al. (3) have synthesized a number of derivatives of arabinosylcytosine, a nucleoside with a very short half-life in vivo because of enzymatic deamination to arabinosyluracil (6). The same problem of rapid deamination pertained to ara-A3 (8). ara-6-MP represented an

Synthesis and evaluation of 2'-substituted cyclobutyl nucleosides and nucleotides as potential anti-HIV agents.

A series of 2'-substituted cyclobutyl nucleoside analogs were efficiently prepared by constructing the core cyclobutyl ring using different [2+2] cycloaddition approaches. The triphosphate derivative of a cyclobutyl nucleoside was also synthesized and evaluated against wild-type and mutant HIV reverse transcriptases (RT). Whereas the nucleoside analogs were inactive against HIV-1 in culture, th...

Interaction of tubulin with ribose-modified analogs of GTP and GDP: evidence for two mutually exclusive exchangeable nucleotide binding sites.

Interactions of tubulin with a number of guanine nucleotides modified at the 2' and 3' ribose hydroxyls were examined. Deoxy analogs of GTP were equal or superior to GTP in supporting tubulin polymerization, but analogs bearing either methyl or phosphate groups on the hydroxyls had significantly reduced ability to support polymerization. These substituted GTP analogs were hydrolyzed at the 5'-g...

Novel acyclic nucleoside analogues as inhibitors of HIV-1 RT

Background Nucleoside reverse transcriptase inhibitors (NRTIs) were the first drugs introduced for treatment of human immunodeficiency virus-1 (HIV-1) infection. These NRTIs may be cyclic or acyclic analogs of natural nucleosides. Both these analogs interact at active site on HIV-RT and compete with indigenous nucleosides/ nucleotides, and thus, divert enzyme activity in manmade direction. All ...

Nucleoside diphosphate kinase and the activation of antiviral phosphonate analogs of nucleotides: binding mode and phosphorylation of tenofovir derivatives.

Tenofovir is an acyclic phosphonate analog of deoxyadenylate used in AIDS and hepatitis B therapy. We find that tenofovir diphosphate, its active form, can be produced by human nucleoside diphosphate kinase (NDPK), but with low efficiency, and that creatine kinase is significantly more active. The 1.65 A x-ray structure of NDPK in complex with tenofovir mono- and diphosphate shows that the anal...

Characterization of the transport of nucleoside analog drugs by the human multidrug resistance proteins MRP4 and MRP5.

The human multidrug resistance proteins MRP4 and MRP5 are organic anion transporters that have the unusual ability to transport cyclic nucleotides and some nucleoside monophosphate analogs. Base and nucleoside analogs used in the chemotherapy of cancer and viral infections are potential substrates. To assess the possible contribution of MRP4 and MRP5 to resistance against these drugs, we have i...
